Background
Triclosan (TCS) and Triclocarban (TCC) are estrogen-mimicking synthetic antimicrobials widely used in personal care products. The massive consumption and associated environmental persistence of TCS and TCC has raised some concerns in terms of their potentially health effects. However, the epidemiological evidence is currently sparse.
Methods
Here, a nested case-control study was performed to prospectively evaluate the association between the exposure of TCS and TCC and the incidence of esophageal squamous cell carcinoma (ESCC). A total of 351 sex- and age-matched participants were followed up and included to document their ESCC occurrence. Detailed epidemiological information was collected at the baseline and the serum levels of TCS and TCC were measured by high-resolution liquid chromatography-mass spectrometry. The association was assessed by multivariable logistic regression, followed by subgroup analyses and sensitivity analyses.
Results
We found a non-linear relationship between the exposure of TCC and the risk of ESCC, of which the fully adjusted odd ratio (OR) of 1.73-7.21 ng/L TCC exposure was 3.46 (95% CI: 1.48-8.07) as compared to the <1.73 ng/L. The exposure of TCS was relatively safe irrespective of their similar structure, indicating their different mechanisms. Table: 405P

Conclusions
Our study revealed a new risk factor of ESCC and alerted the adverse health effect of TCC exposure, contributing to the restriction usage standards of TCC in personal care products.
