Abstract

Aim

The aim of this study was to investigate the incidence of lymphotropic herpes viruses infection in tissue of malignant tumors of epithelial origin.

Methods

Tissue samples of vulvar, tongue and stomach cancer were examined. Reagent kits "QIAamp DNA FFPE Tissue Kit" (QIAGEN, Germany) and MagNa Pure Compact Nucleic Acid Isolation Kit I-Large Volume, (Roche) were used to isolate viral DNA. DNAs of Epstein-Barr virus (EBV) and human herpes virus type 6 (HHV6) were determined by real-time PCR method. 73 tissue samples of vulvar cancer, 54 of tongue cancer, 24 of stomach cancer were studied.

Results

EBV DNA was identified in 16.4% of vulvar cancer tissue samples, HHV type 6 in 8.2 %, in tongue cancer tissue - in 74.1% and 35.7%, in the tissue of gastric cancer - in 91.7 % and 66.7 %, respectively . The frequency of the simultaneous detection of EBV DNA and HHV6 for vulvar cancer tissue was 4.1 %, tongue - 26.2%, stomach - 66.7%. Maximum viral load in tissues of vulvar cancer by EBV was 1200 copies per 100000 cells, the average one - 290 copies. By HHV 6, the figures were 6897 and 1013, respectively. In tongue cancer tissues EBV DNA amount averaged 235 copies per 100000 cells, except for one sample in which the figure was 9484 copies. By HHV6 maximum viral load was 512 copies per 100000 cells, the average - 124 copies. In gastric cancer tissue maximum load amounted to 7890281 copies EBV per 100000 cells, 906 copies by HHV6, the average one - 428 097 and 130 copies per 100000 cells, respectively. The percentage of samples in which EBV DNA amount exceeded 2000 per 100000 cells was 22.7%.

Conclusions

Significant fluctuations in the frequency of infection of cancer tissue of epithelial origin of various localizations with lymphotropic herpes viruses were detected. Genetic material of EBV and HHV6 was mostly detected in tissues of gastric cancer. A higher viral load was recorded in these samples. Studying the role of lymphotropic viruses in the carcinogenesis of tumors of epithelial origin continues.
