Abstract:

Adenoid cystic carcinoma is a rare tumour arising primarily in the salivary gland but can arise in multiple body sites. It is characterized by an indolent clinical course with multiple local recurrences, with overall poor outcome. There has been substantial recent progress defining the molecular underpinnings of this disease. The identification of a recurrent translocation t(6;9)(q22-23;p23-24) resulting in the fusion of the MYB gene on chromosome 6 to the NFIB gene on chromosome 9 in the majority of cases provided a major leap in the characterisation of this rare cancer. We and others have recently applied next-generation sequencing approaches to the disease, characterising mutations primarily via whole exome sequencing. These efforts have led to the identification of mutations which recurrently affect several key pathways including NOTCH signaling, PI3K signaling, chromatin remodeling and growth factor receptor signaling. This work will be presented in the context of potential avenues for therapeutic intervention.
